BD Adds Key P450 and UGT Allelic Variant Human Liver Microsomes to ADMET Portfolio

BD Biosciences – Discovery Labware now provides fully characterized donor panels of P450 and UGT Allelic Variant Human Liver Microsomes (HLM). These HLM panels address the pharmaceutical industry’s research focus in “personalized medicine” and the need to better understand the differences in sub-populations with metabolic allelic variants.

Variability amongst the individual populations as to the efficacy and toxicity of drugs, often results in unpredictable drug responses in patients, and is frequently associated with polymorphisms or genetic variants in genes encoding P450 and UGT drug-metabolizing enzymes. Human Cytochrome P450s and UDP-glucuronosyl-transferases (UGTs) exhibit a large number of allelic variants that may encode for defective enzymes or no enzyme at all. Patients with diminished CYP or UGT activity may require lower than normal doses of specific drugs. For example 3-5% of Caucasians are poor metabolizers of 2C19 and 15% of Caucasians have UGT 1A1(*28*28) a functional UGT promoter polymorphism associated with Gilberts disease (jaundice). Understanding these allelic variants and how individuals metabolize drug candidates differently due to P450 polymorphisms holds the key to prescribing and developing drugs and dosages that are safer for a given patient.
